Intermembrane rhomboid protease activity of Rothia mucilaginosa

چکیده
Rothia mucilaginosa is a part of the normal oral microflora, which has been recognized as an endodontic pathogen and also emerging opportunistic in immune compromised hosts. In this study, we identified rhomboid protease R. candidate virulence factor. A database search two genes on genome with open reading frames 828 bp 822 bp, respectively. The are separated by 57 suggesting that they constitute bicistronic operon. Motifs conserved among family proteins (HxxxN, GxSG, GxxxG) were found their deduced amino acid sequences. recombinant rhomboids rhomboid/liposome complexes (proteoliposomes) prepared using wheatgerm cell-free translation system, serine activities assessed. showed activity form complex, while alone little or no activity. On supposition host invasion, was cultivated medium type I collagen, mRNA levels examined. Real-time reverse transcription polymerase chain reaction analysis revealed mRNAs increased slightly culture although changes not statistically significant. These results suggested possesses active membrane, may be factors involved response to environmental conditions.

Rothia mucilaginosa pneumonia in an immunocompetent patient.
Rothia mucilaginosa is a gram-postive coccus that occurs as part of the normal flora of the oropharynx and upper respiratory tract. Lower respiratory tract infections caused by this organism are rare and usually occur in immunocompromised patients. This is the case of an immunocompetent 47-year-old woman with right upper lobe pneumonia in which R.mucilaginosa was isolated in sputum and bronchia...

The impaired mucociliary clearance in individuals with Cystic Fibrosis (CF) enables opportunistic pathogens to colonize CF lungs. Here we show that Rothia mucilaginosa is a common CF opportunist that was present in 83% of our patient cohort, almost as prevalent as Pseudomonas aeruginosa (89%). Sequencing of lung microbial metagenomes identified unique R. mucilaginosa strains in each patient, pr...

Rothia muciliaginosa (R. mucilaginosa) is a facultative, Gram-positive coccus that is considered to be part of the normal flora of the mouth and respiratory tract. There are sporadic reports of the organism causing endocarditis in patients with heart valve abnormalities, as well as meningitis, septicemia, and pneumonia associated with intravenous drug abuse. However, it is an unusual pathogen i...
متن کاملLate prosthetic joint infection due to Rothia mucilaginosa.
We report a chronic hip arthroplasty infection with Rothia mucilaginosa, a Gram-positive germ belonging to the normal flora of the human oral cavity. Successful treatment was achieved by a two-stage hip arthroplasty revision and intravenous administration of vancomycin. This case report illustrates the potential virulence of R. mucilaginosa in patients with a joint prosthesis. We propose to rou...
